Proteasome inhibitors are drugs that block the proteasome, a cellular protein complex responsible for degrading damaged or unwanted proteins. Inhibiting it causes abnormal proteins to accumulate, which can trigger cell-cycle arrest and apoptosis. Cancer cells—especially plasma cells—can be particularly sensitive to this mechanism.
